DƯỢC LỰC HỌC
– Codein: c6 tac dung giảm ho do tác dụng trực tiếp lên trung tâm gây ho ởhành não.
– Glyceryl guaiacolat: c6 tac dung long dam. Lam giam tính dính của đàm do tăng thể tích
và hàm lượng nước của chất tiết phế quan, vì vậy lam dé khac dam.

Codein: được hấp thu ởruột. Sau khi uống, thời gian bán hủy là 2-4giờ, tác dụng giảm
ho xuất hiện trong vòng |-2giờ vàcó thể kéo dài 4-6giờ. Codein được chuyển hóa ở
gan và thải trừ ởthận dưới dạng tự do hoặc kết hợp với acid glucuronic. Codein hoặc sản
phẩm chuyển hóa bài tiết qua phân rất ít. Codein qua được nhau thai và một lượng nhỏ
qua
được hàng rào máu não.
– Glyceryl guaiacolat: được hấp thu dễ dàng ở ống tiêu hóa, bài tiết qua nước tiểu. Thời
gian bán hủy là Igiờ.
CHỈ ĐỊNH
Làm giảm triệu chứng ho và giúp long đàm
LIEU LUQNG VA CACH DUNG
Người lớn và trẻ em > 12 tuổi: 1viên x1-3 lần /ngày.
Không sử dụng quá 7ngày.
CHÓNG CHÍ ĐỊNH
– Man cam voi bắt kỳ thành phần nào của thuốc.
-_ Trẻ em < 12 tuổi. - Suy hd hap. - Hodo suyén. - Khéng ding dé giảm ho trong các bệnh nung mủ phổi. phế quản khi cần khạc đờm mủ. Phụ nữ mang thai và cho con bú. THẬN TRỌNG -_ Các bệnh đường hô hấp như hen, khí phế thũng. Suy giảm chức năng gan, thận. Có tiền sử nghiện thuốc. -_ Đối với bệnh nhân tiểu đường, cao huyết áp, bệnh tim mạch, rối loạn chức năng thượng thận hoặc giáp trạng, phì đại tiền liệt tuyến, tăng áp lực sọ não. - Than trong đốivới người đang lái xe và vận hành máy móc vì thuốc có thể gây buồn ngủ. TƯƠNG TÁC THUÓC -_ Không dùng rượu trong thời gian điều trị. - Than trong khi phối hợp với Phenothiazine, Barbiturate, Benzodiazepine, IMAO, thuốc chống trầm cảm 3vòng, các dẫn xuất khác của morphin. 1⁄2 - Codein lam giam chuyén héa cyclosporin do tre ché men cytochrom P450. SỬ DỤNG QUA LIEU r9 Triệu chứng: suy hô hấp (giảm nhịp thở, hô hấp Cheyne - Stokes, xanh tím). Lơ mơ dẫn đến == trạng thái đờ đẫn hoặc hôn mê, mêm cơ, da lạnh và âm, đôi khi mạch chậm và hạ huyết áp. Trong trường hợp nặng: ngừng thở, trụy mạch, ngừng tim và có thê tử yong. Xử trí: Phải hồi phục hô hấp bằng cách cung cấp dưỡng khí và hô hấp hỗ trợ có kiểm soát. Chỉ định naloxon ngay bằng đường tiêm tĩnh mạch trong trường hợp nặng. BẢO QUẢN: Bảo quản trong hộp kín, ởnhiệt độ dưới 300C, tránh ánh sáng. DOSAGE FORM: Soft capsule. PHARMACOLOGY - Codeine suppresses the cough reflex by a direct effect on the cough center in the medulla. - Glyceryl guaiacolat has an expectorant action. It is thought to reduce sputum viscosity by increasing the volume and water content of the bronchial secretions. Thereby facilitating the expectoration of sputum. PHARMACOKINETICS - Codeine is absorbed from the gastrointestinal tract. Following oral administration, the half-life of codeine in plasma is 2to 4hours, antitussive effect usually occurs within 1—2 hours and may persist for 4— 6 hours. Codeine is metabolized in the liver and excreted chiefly in the urine as norcodeine and free and conjugated with glucuronic acid. Negligible amounts of codeine and its metabolites are found in the feces. Codeine crosses the placental barrier and is excreted in breast milk. - Glyceryl guaiacolate is well absorbed from the gastrointestinal tract and excreted in the urine. The plasma half-life of glyceryl guaiacolate is 1hour. INDICATIONS Relieves cough and help expectorant. DOSAGE AND ADMINISTRATION Adults and children > 12 years old: 1capsule x 1-3times/day.
Do not use more than 7 days.
CONTRAINDICATIONS
– Patients with hypersensitivity to any component of this medicine.
– Children < 12 years old. - Ventilatory failure. - Asthmatic cough. - Do not use to relieve cough to people with lung abcess, bronchitis which need to cough to expel purulent sputum. - Pregnancy and lactation. PRECAUTIONS - Respiratory diseases including asthma, emphysema. Hepatic and renal impairment. History of drug abuse. - Patients with diabetes, hypertension, cardiovascular disease, adrenal dysfunction or thyroid, prostate enlargement, increased cranial pressure. - Patients should be cautioned about driving and operating machinery because the drug may cause drowsiness. 1/2 ae - DRUG INTERACTIONS - Do not drink alcohol during treatment. - Caution should be combined with phenothiazines, barbiturates, benzodiazepines, MAO inhibitors, tricyclic antidepressants, other derivatives of morphine. - Codeine reduce metabolism of cyclosporine, due to inhibition of cytochrome P450. PREGNANCY AND LACTATION Do not use this drug for pregnant women and lactating. EFFECTS ON ABILITY TO DRIVE AND OPERATE MACHINES Patients should be cautioned about driving and operating machinery because the drug may cause drowsiness. SIDE EFFECTS Drowsiness, nausea, vomiting, constipation, dizziness, dazzle, rash may occur. Inform your doctor in case of any adverse reactions related to drug use. OVERDOSE Symptoms: Respiratory depression (a decrease in respiratory rate or vital volume, Cheyne- Stokes respiration, cyanosis). Extreme somnolence progressing to stupor or coma, miosis, skeletal muscle flaccidity, cold and clammy skin, and sometimes bradycardia and hypotension. In severe overdose, apnoea, circulatory collapse, cardiac arrest and death may occur. Treatment: Primary attention should be given to the reestablishment of adequate respiratory exchange through provision of apatent airway and the institution of assisted or controlled ventilation as necessary. Naloxone may be administered, preferably by intravenous routed in the severe patient of overdose. STORAGE: Stored in ahermetic container, below 30°C, protect from light.
